Manga ini bercerita mengenai Death Note (buku kematian) yang sengaja dijatuhkan oleh Ryukk, seorang Shinigami (malaikat kematian, reaper) ke dunia manusia, di mana bila nama seseorang ditulis dalam buku tersebut, maka orang itu akan segera meninggal. Kematian para kriminal yang tidak wajar dan dalam waktu yang hampir bersamaan ini membuat masyarakat dan pihak kepolisian merasa kejadian ini bukanlah terjadi secara kebetulan. Meskipun terdengar tidak masuk akal, pihak kepolisian mulai merasa ada seseorang di balik semua kejadian yang menimpa para kriminal tersebut. Raito yang menggunakan kekuatan Death Note kini disebut sebagai Kira (Killer dalam dialek jepang) dan dianggap sebagai dewa oleh orang-orang yang pro dengan tindakan Raito tersebut.
